Funding Opportunity Purpose

This Funding Opportunity Announcement (FOA), issued by the National Institute on Aging (NIA) at the National Institutes of Health, solicits grant applications that propose extension of research activities on osteoimmunology in order to broaden our understanding of aging aspects of osteoimmunology, using animal models. Most studies in this field have been conducted in young mouse models (usually animals less than 6 months of age). Under those conditions, it is not possible to ascertain the effects of aging in the balance between bone metabolism and immune function. Studies sought under this FOA should focus on the impact of the age-related decline in immune function on bone homeostasis or conversely on the age-related changes in the bone marrow niche affecting the immune interactions with bone. Research in osteoimmunology aims to understand the interplay between the immune and skeletal systems, and how these interactions lead to disease when disrupted. In the clinical setting, such disequilibria and the resulting diseases are most commonly found in the elderly, which are afflicted by both immune senescence and loss of bone homeostasis. This FOA is intended as a vehicle to allow investigators already working in the field of osteoimmunology to expand the scope of their research, so as to allow an enhanced understanding of the field within the context of the clinically relevant setting of aging.

Section I. Funding Opportunity Description

Purpose

The purpose of this Funding Opportunity Announcement (FOA) is to encourage scientific collaborations between investigators with expertise in osteoimmunology and aging biology in order to discover mechanisms modulating age-related changes in the interactions between the immune system and bone.

Background

A decline in immune function with aging has been known for some time and many alterations in the function of both the adaptive and innate immune systems have been documented. The knowledge that immune cells are mobilized from bone marrow led to the demonstration that different types of immune cells interact with osteoblasts and osteoclasts, and these interactions have subsequently been shown to modulate bone homeostasis.

Osteoimmunology and Aging

Bone is continually adapting to physiological changes in order to preserve skeletal size, shape, structural integrity and mineral homeostasis via integrated cross talk with multiple tissues. Recent reports from multiple laboratories indicate a role of senescent T cells in the development of osteoporosis. Unexpectedly, mice lacking either B- or T-cells have osteoporotic bones, suggesting that these immune cells participate in the maintenance of bone homeostasis during basal physiology. These observations have led to a growing interest in the field of osteoimmunology, i.e., the interactions between these two physiological systems and how changes in immune function might affect bone physiology. However, while the main changes in immune function that are relevant to bone health are those that occur as a function of age, to date most studies in osteoimmunology have used mice in what would otherwise be a healthy young adult animal (3-6 months of age). However the field has not systematically addressed the mechanisms by which the decline in immune function and hematopoiesis with age affects maintenance of normal bone remodeling and metabolism.

Parabiotic experiments in the field of aging have clearly demonstrated that the aged extracellular niche is substantially different from that of a young adult in terms of the ability to repair damage and heal normally; both of these are immune mediated functions. Data demonstrating central and endocrine regulation of both the immune and skeletal systems indicate a strong scientific basis and growing need to further investigate how aging impacts the mechanisms involved in mediating cross talk between these systems. Moreover, numerous recent studies have indicated a central role for bone cells in the regulation of hematopoiesis and metabolism. Hematopoietic stem cells (HSCs) generate all of the hematopoietic lineages and are capable of self-renewal, and it is known that aging has a profound effect on these functions leading to impaired lymphopoiesis (skewing of the immune cell population) and enhanced myelopoiesis (increasing the number of bone resorbing osteoclasts). Stem cell intrinsic factors and extrinsic factors associated with the aged microenvironment may contribute to the overall decline in function of the hematopoietic system and a better understanding of the mechanisms mediating how aging affects hematopoietic differentiation is essential to delineating the physiological integration of the immune and skeletal systems. Age-dependent changes in immune cell recruitment and differentiation will have a significant effect on the ability to recruit and differentiate appropriate numbers of osteoblastic and osteoclastic precursor cells.

Research Objectives

It has been recognized by both immunologists and bone biologists that a more complete understanding of the role of the immune system in regulating skeletal homeostasis during aging is required. Aging studies constitute a vital and largely unexplored research area ready for further investigations in this field.

In view of this need, the NIA is issuing this FOA to encourage studies on aging in the field of osteoimmunology. Studies examining the interactions of the immune and skeletal systems with advancing age using state-of-the-art techniques and approaches are encouraged.

For the purpose of this FOA, the main interests are in cellular and molecular mechanisms in established models of osteoimmunology that have been previously or are currently funded. Areas of interest and appropriate topics in aging research include, but are not limited to:

It is expected that applications in response to this FOA will have an existing established, or previously funded model of osteoimmunology. The differences in experimental approaches in these fields are viewed as complementary and essential for a more complete understanding of the role of aging in immune skeletal interactions. Research applications intended to study the role of the immune system in bone homeostasis but not investigating aging in modulating this interaction will not be considered responsive to this FOA. Part of the experimental approach is expected to include testing endpoints at multiple ages including 24 months of age. The use of animal models that have reached advanced ages will be required. Potential applicants are encouraged to discuss their scientific interests with the scientific staff mentioned in Section VII when planning the application.

NIA intends to fund between 4-5 awards, corresponding to a total of $1 million for fiscal year 2013. Future year amounts will depend on annual appropriations.

Section V. Application Review Information

1. Criteria

Only the review criteria described below will be considered in the review process. As part of the NIH mission, all applications submitted to the NIH in support of biomedical and behavioral research are evaluated for scientific and technical merit through the NIH peer review system.

Overall Impact

Reviewers will provide an overall impact score to reflect their assessment of the likelihood for the project to exert a sustained, powerful influence on the research field(s) involved, in consideration of the following review criteria and additional review criteria (as applicable for the project proposed).

Scored Review Criteria

Reviewers will consider each of the review criteria below in the determination of scientific merit, and give a separate score for each. An application does not need to be strong in all categories to be judged likely to have major scientific impact. For example, a project that by its nature is not innovative may be essential to advance a field.

Significance

Does the project address an important problem or a critical barrier to progress in the field? If the aims of the project are achieved, how will scientific knowledge, technical capability, and/or clinical practice be improved? How will successful completion of the aims change the concepts, methods, technologies, treatments, services, or preventative interventions that drive this field?

Investigator(s)

Are the PD(s)/PI(s), collaborators, and other researchers well suited to the project? If Early Stage Investigators or New Investigators, or in the early stages of independent careers, do they have appropriate experience and training? If established, have they demonstrated an ongoing record of accomplishments that have advanced their field(s)? If the project is collaborative or multi-PD/PI, do the investigators have complementary and integrated expertise; are their leadership approach, governance and organizational structure appropriate for the project? Does the team show appropriate experience in aging research?

Innovation

Does the application challenge and seek to shift current research or clinical practice paradigms by utilizing novel theoretical concepts, approaches or methodologies, instrumentation, or interventions? Are the concepts, approaches or methodologies, instrumentation, or interventions novel to one field of research or novel in a broad sense? Is a refinement, improvement, or new application of theoretical concepts, approaches or methodologies, instrumentation, or interventions proposed?

Approach

Are the overall strategy, methodology, and analyses well-reasoned and appropriate to accomplish the specific aims of the project? Are potential problems, alternative strategies, and benchmarks for success presented? If the project is in the early stages of development, will the strategy establish feasibility and will particularly risky aspects be managed? 

If the project involves clinical research, are the plans for 1) protection of human subjects from research risks, and 2) inclusion of minorities and members of both sexes/genders, as well as the inclusion of children, justified in terms of the scientific goals and research strategy proposed?

Environment

Will the scientific environment in which the work will be done contribute to the probability of success? Are the institutional support, equipment and other physical resources available to the investigators adequate for the project proposed? Will the project benefit from unique features of the scientific environment, subject populations, or collaborative arrangements?

Additional Review Criteria

As applicable for the project proposed, reviewers will evaluate the following additional items while determining scientific and technical merit, and in providing an overall impact score, but will not give separate scores for these items.

Protections for Human Subjects

For research that involves human subjects but does not involve one of the six categories of research that are exempt under 45 CFR Part 46, the committee will evaluate the justification for involvement of human subjects and the proposed protections from research risk relating to their participation according to the following five review criteria: 1) risk to subjects, 2) adequacy of protection against risks, 3) potential benefits to the subjects and others, 4) importance of the knowledge to be gained, and 5) data and safety monitoring for clinical trials.

For research that involves human subjects and meets the criteria for one or more of the six categories of research that are exempt under 45 CFR Part 46, the committee will evaluate: 1) the justification for the exemption, 2) human subjects involvement and characteristics, and 3) sources of materials. Inclusion of Women, Minorities, and Children 

When the proposed project involves clinical research, the committee will evaluate the proposed plans for inclusion of minorities and members of both genders, as well as the inclusion of children. Vertebrate Animals

The committee will evaluate the involvement of live vertebrate animals as part of the scientific assessment according to the following five points: 1) proposed use of the animals, and species, strains, ages, sex, and numbers to be used; 2) justifications for the use of animals and for the appropriateness of the species and numbers proposed; 3) adequacy of veterinary care; 4) procedures for limiting discomfort, distress, pain and injury to that which is unavoidable in the conduct of scientifically sound research including the use of analgesic, anesthetic, and tranquilizing drugs and/or comfortable restraining devices; and 5) methods of euthanasia and reason for selection if not consistent with the AVMA Guidelines on Euthanasia. 2. Review and Selection Process

Section VI. Award Administration Information

1. Award Notices

If the application is under consideration for funding, NIH will request "just-in-time" information from the applicant as described in the NIH Grants Policy Statement.

A formal notification in the form of a Notice of Award (NoA) will be provided to the applicant organization for successful applications. The NoA signed by the grants management officer is the authorizing document and will be sent via email to the grantee’s business official.

Awardees must comply with any funding restrictions described in Section IV.5. Funding Restrictions. Selection of an application for award is not an authorization to begin performance. Any costs incurred before receipt of the NoA are at the recipient's risk. These costs may be reimbursed only to the extent considered allowable pre-award costs.

When multiple years are involved, awardees will be required to submit the annual Non-Competing Progress Report (PHS 2590 or RPPR) and financial statements as required in the NIH Grants Policy Statement.

A final progress report, invention statement, and the expenditure data portion of the Federal Financial Report are required for closeout of an award, as described in the NIH Grants Policy Statement.
